Determination of 9 kinds of water-soluble vitamins in sports drinks by liquid chromatography-mass spectrometry

摘要：建立液相色谱-质谱法快速测定运动饮料中维生素B1、维生素B2、泛酸、维生素B6、生物素、叶酸、维生素B12、烟酸、烟酰胺9种水溶性维生素的方法。样品用水超声提取后，采用ACQUITY HSS T3液相色谱柱分离，以0.1%甲酸甲醇-0.1%甲酸水溶液为流动相，梯度洗脱，采用质谱多反应监测扫描模式的选择性和特异性定量。9种水溶性维生素的质量浓度与色谱峰面积分别在各自范围内具有良好的线性关系，线性相关系数均大于0.999。当称样量为2.00 g，定容体积为50 mL时，方法检出限为0.025 0～0.062 5 μg/g，测定下限为0.10～0.25 μg/g。样品的加标回收率为90.0%～102.3%，测定结果的相对标准偏差为0.9%～3.2%(n=7)。该方法简便、快速、准确、灵敏，适用于运动饮料中水溶性维生素的测定。
Abstract：A method for the rapid determination of 9 kinds of water-soluble vitamins in sports drinks by liquid chromatography-mass spectrometry(LC-MS) was established, including vitamin B1, vitamin B2, pantothenic acid, vitamin B6, biotin, folic acid, vitamin B12, niacin and niacin amide. The samples were extracted with water and separated by ACQUITY HSS T3 liquid chromatography column. Gradient elution was carried out by using 0.1% formic acid methanol-0.1% formic acid aqueous solution as mobile phase. The selectivity and specificity quantitative of mass spectrometry multiple reaction monitoring(MRM) scan mode were selected. The mass concentration of the 9 kinds of water-soluble vitamins had a good linear relationship with the chromatographic peak areas in its own linear range, and the correlation coefficients were more than 0.999. As the sample volume was 2.00 g and the constant volume was 50 mL, the detection limits were 0.025 0-0.062 5 μg/g, and the detection limits were 0.10-0.25 μg/g. The recoveries of the samples were 90.0%-102.3%, and the relative standard deviations were 0.9%-3.2%(n=7). The method is simple, rapid, accurate and sensitive, and it is suitable for the determination of water soluble vitamins in sports drinks.
