摘要：建立高效液相色谱法同时测定苹果中D-果糖、D-葡萄糖和蔗糖含量的方法，并对不同产地样品中3种糖进行主成分分析和聚类分析，为苹果质量评价与分类提供依据。用单因素实验对苹果样品前处理条件进行优化，确定了最佳样品前处理条件：离心转速为16 000 r/min，离心时间为17 min，料液比为1∶12。D-果糖、D-葡萄糖和蔗糖的线性范围分别为0.245～1.495 mg/mL(r=0.999 1)，0.104～1.004 mg/mL(r=0.999 6)，0.075～1.025 mg/mL (r=0.998 2)。果糖、葡萄糖、蔗糖的加标回收率分别为96.83%～100.08%，83.51%～96.40%，93.97%～98.89%，测定结果的相对标准偏差为0.69%～1.17%(n=6)。通过聚类分析将6个产地的苹果样品分成了3类，与主成分分析结果相同。该方法简便、准确，可用于测定苹果果实中糖的类型及其含量，为苹果品质鉴定提供科学依据。
Abstract：The method for simultaneously determining D-fructose, D-glucose and sucrose in apple by HPLC-ELSD was established and these water-soluble sugar constituents in different growing areas were carried out by principal component analysis and cluster analysis in order to provide basis for evaluating its quality and classification. Single-factor experiment was used to optimized the pre-processing conditions of apple sample, the optimal sample pretreatment conditions were followed: the centrifugal speed was 16 000 r/min, the centrifugal time was 17 min, the solid-liquid ratio was 1∶12. The linear ranges of D-fructose, D-glucose and sucrose were 0.245-1.495 mg/mL(r=0.999 1), 0.104-1.004 mg/mL(r=0.999 6) and 0.075-1.025 mg/mL (r=0.998 2), respectively. The recoveries of D-fructose, D-glucose and sucrose were 96.83%-100.08%, 83.51%-96.40% and 93.97%-98.89%, respectively, and the relative standard deviations of determination results were 0.69%-1.17%(n=6). Cluster analysis was applied to divide samples from 6 different habits into 3 categories with the same as the result of principal component analysis. This method was simple and accurate, and can be used to determine the type and content of water-soluble sugar in apple, it can provide a scientific basis for apple quality identification.
